The bachelor thesis is focused on new ways of using raw materials in the brewing in-dustry and new innovations. The theoretical part mentions basic raw materials for beer production, special types of malt, solid adjuncts from other raw materials, especially pseudocereals, liquid adjuncts, various methods of processing hops and new proce-dures or new equipment in beer production. Also the principle of production of Czech beer and European beer is compared. In the second part, the possibility of using star-chy surrogates was investigated. A recipe was created using buckwheat and then the sensory profile of the beer was created. From the obtained results, it was found that the addition of buckwheat negatively affected the sensory properties of the beer, which was reflected in the negative evaluation of both aroma and taste.
Development of fortified gluten-free cereals for specific nutrition

The bachelor thesis was focused on the development of enriched gluten-free cereals for special nutrition. The fortified cereal product included processed microbial biomass, either as a freeze-dried powder or as an extract, for which the phenolic content, antioxidant content and protein content were determined. The theoretical part includes findings from professional literature on gluten-free cereals, pseudocereals and their chemical composition. Subsequently, a review on microalgae and yeasts was prepared including a list of their metabolites. The literature search focused on green microalgae and carotenogenic yeasts. In the experimental part, the characterization of the selected pseudocereals and optimization of the prepared extracts of the selected microalgae and carotenogenic yeast were firstly carried out. The selected pseudocereals were amaranth, buckwheat and quinoa. The green microalgae strain Chlamydomonas reinhardtii and the carotenogenic yeast strain Cystofilobasidium macerans were selected for enrichment of the pseudocereals. Carbohydrates, proteins, fatty acids, -glucans, gluten, vitamins, phenolic substances, flavonoids and antioxidants were determined in the pseudocereals. Total fatty acids, lipophilic substances, phenolic substances and total antioxidant activity were detected in the microbial biomass. Finally, mixed cereal products were prepared from pseudocereal flour, microbial biomass and microbial extract. The cytotoxicity of the selected extracts was determined by MTT assay. It was found that the addition of algal biomass alone or algal extract to cereal flours resulted in a significant increase in protein. The yeast ethanol extract increased the phenolic and antioxidant content of the cereal products.

The literary part of the diploma thesis deals with traditional raw ma-terials, special malts and non-traditional raw materials for beer producti-on and beer production as such. Furthermore, the production of beers from non-traditional raw materials in the world. The experimental part is devoted to the description of the production of own beers from non-traditional malts (amaranth, chickpeas, sorghum, buckwheat, quinoa) and their chemical analysis by HPLC and Fermen-toFlash. In the next part a method of sensory analysis was described, and the results were then shown using radar charts. In the results and discussion section these analyzes are statistically processed and evaluated for the mutual dependence of the substances from the chemical analysis on the sensory characteristics of the individu-al products.

The bachelor thesis Buckwheat in Food Technology is focuses on a relatively popular and richly grown crop, it is buckwheat. Pseudo-cereal, which ranks to grain for her properties, is again on the rise after a long recession and enjoying rising popularity. The work describes origin, history of use, botanical characteristics and cultivation. To a great extent, I also focus on the chemical composition of buckwheat coatings, especially with regard to the substances most frequently found in this plant, which are very valuable to us. The next part describes technological processing in mills and bakeries. Last but not least, is mention non-food use as livestock feed and pillowcases.

The effect of pyrazine derivatives on the secondary metabolites production in in vitro cultures of medicinal plants I.

The aim of this work was to investigate the effect of pyrazine derivative, 1-octyl-3-(pyrazin-2- yl)urea, as an abiotic elicitor on the production of flavonoid rutin in in vitro cultures of Fagopyrum esculentum Moench., Cultivar Bamby. Suspension and callus cultures were cultivated on Murashige and Skoog nutrient medium (MS) with the addition of 2,4-dichlorophenoxyacetic acid (2,4-D) as growth regulator at a concentration of 1 mg/l. The elicitor solution was added to the cultures at three concentrations: c1 (100,0 mg/100 ml), c2 (10,0 mg/100 ml) and c3 (1,0 mg/100 ml). The elicitor was monitored at six time intervals: 6, 12, 24, 48, 72 and 168 hours. To control samples 1 ml of ethanol 96% was added instead of elicitor solution and samples were collected after 24 and 168 hours. Samples were taken at given time intervals and dried. Subsequently, the rutin content was monitored by HPLC. The rutin release into the nutrient medium was also tested. During the experiment on the callus cultures no statistically significant increase in rutin production after elicitor treatment was observed. But elicitor increased rutin production in suspension cultures after treatment in all tested concentrations. The calluses always released rutin into the nutrient medium. Evaluation of functional properties of selected vegetable flours

This bachelor thesis deals with basic structure, nutritional properties, production and use of flours. There are information about cereals and legumes, which are used for productions of flours. This thesis talks about food intolerances, which can be a reason why change basic flours of wheat, barley, oat and rye to another. Practical part is about 14 flours, which were tested on dry matter, humidity, nitrogen substances content, proteins, protein electrophoretic profiles (patterns), solubility, water and fat holding capacity, formation of gels and changes of colours during a boiling. The results of this research are information about nutrition and functional properties of tested flours.

Higher plants represent an important source of valuable substances, so called secondary metabolites, which can be obtained through explant cultures of plants. Elicitation is a method of increasing the secondary metabolites production. This study aims to evaluate the secondary metabolites production in Fagopyrum esculentum variety Spacinska cultures in vitro after abiotic elicitor treatment. The experiment was focused on alteration of rutin production in callus and suspension cultures of F. esculentum var. Spacinska after selenium application. Murashige and Skoog nutrient medium supplemented with 1 mg l-1 2,4-dichlorophenoxyacetic acid was used for the cultivation. Selenium solutions of various concentrations (c1 = 9.012×10-3 mol l-1 , c2 = 9.012×10-4 mol l-1 , c3 = 9.012×10-5 mol l-1 ) were affecting the cultures for 6, 12, 24, 48, 72 and 168 hours. The content of rutin was determined by HPLC. The release of secondary metabolites into the nutrient medium was studied as well. After elicitor application, the rutin production increased in both callus and suspension cultures. Higher levels of rutin content were detected in callus culture.
